So I've been doing some practice on this and while doing I found a small timesaver in the Monument Island Gateway chapter. Not sure if those are already known.
After pulling the lever that makes the cargo move forward during the Skyline section you can jump in front of it. That's already known (not sure if that saves time). However, if you manage to jump off in a way that you will be really close to it when you hit back onto the Skyline you can get a boost that makes you get to the end of that Skyline in like a second. You do have to spam Space in order to jump onto the Freight Hook as soon as possible otherwise you will get stuck in the cargo that hangs there and you have to switch direction to get unstuck. Also if you jump into the moving cargo you also get stuck and have to switch direction. I've been getting it somewhat consistent and I almost never get stuck so this is something trying to go for is worth it.
e: Here's a video showing it off.
The second thing I noticed is that if you throw a Devils Kiss Trap at a specific point of the guardrail of the airship while riding the elevator up the two guards inside won't spawn. After some more testing I just did this seems really inconsistent. I have had it work every time on one reset from checkpoint, but after reloading the entire chapter it only worked once. Sometimes it works and sometimes it doesn't. Here is a screenshot of where I try placing it, and since it does kill the female guard it doesn't hurt trying to go for this either.

Holy Mother of something I think I just managed to skip the Downtown Emporia Siren Sidequest. I'll just upload the footage now and then continue further testing, you don't believe how excited I am right now.
I'll edit the video in in a few minutes.
e: There we go
e2: Video taking a while to process, here's some good news: I just did it again. The third suicide isn't even neccessary. Now the only thing that could potentally screw with this is the fact that I noclip'ed from the Downtown Emporia Chapter to the gate not doing the skip because I'm a scrub, but I don't really see why that would interfere.
e3: It's really weird. I still haven't figured out the exact positioning on step 1 (suicide through gate) and 3 (jumping off the vault wall). Apparently you don't even have to suicide in step 3, you just have to fall correct.
MATHEMATICS
Holy shit, that's awesome news!
WHAT THE EVERLOVING FUCK IS THIS HOLY SHIT NOBODY DO RUNS UNTIL I GET SUB-2:00
Sub 2:00 totally possible now, yeah. I did some rough timing with your 2:09 run and it saves like 9-10 Minutes.

This is based as fuck
Okay so for me, after getting behind it once and then suiciding behind the gate, it doesn't actually matter what I do with the second jump. I respawned behind the gate 3 times in a row, one was a suicide the other two just normal jumps. I basically stood at about the same place on the wall as in my video, turned my back to the void and jumped slightly backwards.
Looks like it does come down to the suicide through the gate. I'm still not sure if you have to touch the ground or a wall or a specific point, or if you just have to get in very far. From my experience it seems the little space between the gate and the pillar get me to the other side, and I also respawned at that place as well as Ferret in his successful attempt iirc.

My suspicion is that the suicide through the gate needs to allow you to touch the ground on the other side (as opposed to a wall, or whatever). I'm downloading the game again so I'll mess with it later tonight/tomorrow.

I MIGHT HAVE A REASON TO RUN THIS GAME AGAIN Cheesy I'll try to test this out on the console version tonight or the next few days
Nice find! I'll add the skip to the doc once there is an optimized clip of the skip. It seems like there is some more testing to do.

Unable to emulate the trick on PS3 thus far. Everything lines up accordingly--it's funny to see the loading ("deloading") screen when you go from the statue/gate area to the second suicide location. It's easy to see why this trick is possible when you can pretty much watch the game unload the area that you then use to get out of bounds. Basically, it doesn't load the area back when it spawns you leaving the big chasms. But anyway, not telling anyone anything they don't already know.

More practically:

I've hit the floor several times in the initial gate suicide. I don't know what the trigger is, but it doesn't seem to be floor contact. I'm going to try some distance jumps/wall hits. I spawn at varying distances between the gate and the statue depending on how far out I jump on the final step (Further jumps = closer to statue, trying to jump close to or under the building gets closer to the gate, but no luck getting anywhere near the other side.)

Basically, this is all about finding the trigger at this point.

EDIT: A couple of times I have spawned right up against the gate but not on the other side. Are we sure the success doesn't depend on how you do the final jump? Looking forward to a clip where it's done without the last suicide.

Great find RealLink!!!
My guesses so far after messing with it:

Only the first two suicides are necessary. The third is definitely useless, and Link has done the skip successfully without the last one, so best guess is that that one doesn't do anything (though it's possible that it helps, I don't know why it would). The important thing about the last jump off the roof of the bank is that you don't get the infinite death glitch, which you avoid by strafing back towards the bank as you fall.

The main factor right now is getting the game to set your respawn location behind the gate. Basically, we want the game to set your respawn location just as you're doing the suicide and land on the other side. The game appears to periodically set your current position as a respawn location, meaning that if you fall onto the death plane, it will respawn you at that location. The question is how the game calculates when it will set a respawn location. It might be on a timer (set new respawn location every X seconds), based on location (set new respawn location when you move X distance from the last respawn location), or based on something else. If this theory is right, we need to figure out how to manipulate the game into setting the respawn location when we want it to - as we do the death warp.

Or maybe all of this is complete BS and I'm just delusional after bashing my head against a wall for hours yesterday.

My worst fears are coming true. Someone tell me I'm wrong. If I'm not, heaven help us all.

I think this is Elizabeth-based.

I've done about 30 jumps worth of testing and every single time, after the final jump, I've ended up in the original location Elizabeth was at right when I did the first suicide at the gate. If you can get her up against the gate, you'll respawn against the gate. If you run really far down the steps toward Victory Square and then make a mad dash for it, you'll respawn on the stairs somewhere, right about where she'd be. It didn't seem very likely at first, but I've now done enough of these and I'm a believer. I think Elizabeth is the key to making this work basically making it Factory Skip 2.

EDIT: GRRRRR RANDOMNESS. The Liz trend has gone away. Good news except that I still can't get a success. DansGame.
I don't really see it; I've done dozens of jumps where Elizabeth was waiting by the gate in front of the handprint thing, and I spawned in many different locations. I don't really see any reason why it would depend on Elizabeth's location, either.
I've gotten random locations the last 2 times by, so yeah, your guess is as good as mine at this point. I thought maybe it was a "last known" type thing, but yeah, it still seemed pretty out there.

Nevertheless, I've gotten up against the gate twice and both times were with Elizabeth standing about where she was in the success video. Aside from that I can't find much consistency.
This is the worst >_< I haven't been able to replicate it after the 5 times I got it yesterday, and FF only got it once. I also thought it would be a combination of timer and distance between the last checkpoint because of how the game handles the respawns when you jump off the map normally without going OOB.
Another idea I had yesterday was forcing the game to set the "Restart from Checkpoint" checkpoint you get from activating the gate for the first time behind the gate, but unfortunately the game only sets that to your current position when you are alive. If you kill yourself anytime after you activate it it sets the checkpoint to the respawn location next to the vending machines and puts you in No-armed-bandit state (no weapon refill, no weapons or vigors until you melee attack or activate another cutscene), which seemed like a completely useless glitch.

I tried the same RealLink. What's worse is that I was out of ammo when I did it so I had the reload prompt taunting me like a chump.

EDIT: Handyman spawn is probably no longer necessary in Emporia Downtown fight. That's a brief time saver. FF also mentioned doing the lift enemy/charge into them over the gate possibility that was previously abandoned because of Handyman spawning necessities.

Stoic_Squirrel got the skip a few times. It's his belief it is all about the suicide position and that the final jump doesn't matter. Also, the 2nd suicide isn't necessary either. (I don't know how many people were doing it to replicate the video exactly.)
Works on PS3 confirmed. Got it 2 out of 3 times by doing jump, rocket, and trying to let Booker land in the middle of the gate. I used Stoic_Squirrel's videos as a guideline.

http://www.twitch.tv/stoic__squirrel/c/3834496

For some goofy reason on mine, I also got the objective message "Go to Lady Comstock Memorial" in just black, without the usual red flag around it. Pretty sure that's just convoluted autosave + glitch nonsense leftover from doing all kinds of goofy shit to that playthrough file. The only advice I have for the moment is to try to land in the center of the gate. I definitely "felt" the second one being a success after doing it once.
Sorry to be stacking posts, but I think this is my new route (From Emporia Fight ----> Comstock Gate Skip):

At Downtown fight, instead of fighting, then waiting for Handyman, I only fight enough to trigger the 2nd barge. Then I do the Elevator jump as normal, but instead of doing the big leap, I go right to the flag stand. Then I jump from there onto the "greenhouse" looking roof. Then I shoot myself down to suicide range, then jump to the right letting the fall kill me. This spawns me where the harder jump is, but it's almost no time difference (On PS3 anyway since there's a load during the difficult usual jump) and it's infinitely safer. It also keeps the guys from spawning in the square just as the typical jump does.

Important note: You do not have to wait for Elizabeth to trigger the "Hello Lady Comstock!" dialogue or even for her to get to the gate. You can just run ahead and do the suicide as soon as you get there.

I now have the trick down. I can do it about 80% of the time and when I miss, I know I miss and can just try again immediately instead of hoping for the best and doing the whole thing.

As soon as you can get us some videos, that would be great.

EDIT: also, Z1m's really old way of getting out of Downtown Emporia fight is now probably the fastest way out.

When I get back into town from work, I can take a crappy vid for educational purposes, but I'm still a month-ish from being able to stream.

I did a few practices this morning and hit 2 out of 5. There was only once where I thought I got it and didn't. It's precise for sure, but given how much time it saves, I think you'd have to screw it up a dozen times or so before you started losing time over the Ghosts fights.
Can you give a really precise description of your setup, then? Do you try to land inside the gate itself? What's your setup for the jump?
Really great shortcut and not so hard to get.

The only catch for me is if you fail it, that's instant reset. If I don't get the suicide through the gate right, when I'm supposed to be loaded behind the gate after jumping down the bank walls, I end up in an infinite death loop (and restarting checkpoint takes you all the way back to the start of emporia fight). And I can't seem to tell when the jump is done right or not.
You can avoid the infinite death loop by making sure the fall off the bank is in the right place; generally, try to strafe back under the bank and you should be fine.
Gets the cake.
