Okay, I think it's time some real discussion starts for this game. NO SUCCUBUS GLITCH!!!!!!

Basiclly what I've came up with is the following. The time to shoot for is under 2 hours, unless something is skippable. No extra souls will be collected except Black Panther (duh).

There will be very frequent visits to Hammer to stock up on mind ups. a bunch of equipment will be picked up, but most of it will not be used. five hidden rooms could be used (ninja suit, satan's ring and fragarach, durandal, olrox's suit, foei gras), some may not be necessary.

the claymore will be used for most of the game until the Durandal comes around. axe armor and killer clown will be the most used souls until Aguni and Abbadon. Use of Backdash Cancelling will be very frequent.

I'll add more later. I'm tired.

However, you'll want to get a backstabbing weapon to open the oneway gates early, which will certainly save a good amount of time. For instance, the golden gate before the entrance to Condemned Tower and the "backdoor" in Subterranean Hell that leads to the Silenced Ruins. Can't think of much else right now. Don't forget that in theory much more sequence breaking would be possible, but you can't fight the bosses without the proper seal.
Of course then comes the issue of someone with....

1. A DS Lite
2. A copy of Castlevania: DoS
3. Willing to speed run it
4. has the "proper" equipment to record it

I'd love to do it, but I have neither the skills or the recording equipment to run it....also, don't forget about the difference in times between New Game, New Game+, New Game+ Hard, Julius Mode, Boss Rush Mode, and Julius Boss Rush Mode
The first run has to be a New Game on Normal Mode.
I tried this a long time ago with a time of 1:07:36 in 14 segments (with backstab to open doors, but no Succubus glitch).  I don't have the means to record it.

Backstab should be used three times: to enter the clock tower immediately after getting the Cutall, to enter Subterranean Hell from the Wizardry Lab, and on the way to the Mine of Judgement.  Entering the Silenced Ruins through the back door is slower, since you will have Zephyr's soul by this time.

Doppelganger: From the moment you get the Cutall until the backstab on the way to the Mine of Judgement, the "B" setup should be as much as you have of Cutall, Mina's Talisman, Bat Company, and Paranoia.  After that backstab, there is no need for any of those, so my B at that point is a copy of my A except for the bullet soul.  This way I can quickly switch between Aguni and Axe Armor while fighting Abbadon.

My route, to the best of my memory:

1. Get initial 3 souls, get Axe Armor soul, kill Flying Armor, upgrade to Cutlass, activate Wizardry Lab warp, kill Balore.
2. Activate garden warp, get Corpseweed soul, get Doppelganger soul.
3. Kill Dimitrii and Malphas.
4. Get Great Armor soul, warp, upgrade Cutlass to Fragarach, warp to garden, get Ninja Suit.
5. Defeat Dario, get talisman, get Lilith soul, get Cutall, immediately head back and backstab into clock tower.
6. Get magic seal 4 (skip seal 3), kill Zephyr, warp to Wizardry Lab, backstab into Subterranean hell, get whatever that armor is on the way there.
7. Get Ukoback soul, kill Bat Company, warp back to clock tower, save in Pinnacle.
8. Get Killer Clown soul, kill Paranoia, get Olrox's Suit, get Mana Prism.
9. Get Durandal, go through the Pinnacle.
10. Get Satan's Ring, kill Aguni, get Truffles, warp to village, upgrade to Dainslef, warp to garden, watch scene, backstab through gate, go through Mine of Judgement.
11. Kill Death.
12. Go through Abyss.
13. Kill Abbadon, get Black Panther soul, save before Menace.
14. Kill Menace.

My route can undoubtedly be improved, but it should be a decent starting point.  I haven't experimented with different weapons or souls to kill bosses more quickly.
Nice route. I'd also recommend getting Barbariccia, it's not as fast as the transformation souls in AoS, but certainly faster than walking, and there are enough long hallways in this game. You won't get black panther until the very end.

Here is a Speedtrick using the succubus glitch:
In the big pinnlace room with the stairs to Aguni, go to the upper right corner when first entering the room and use the succubus glitch with an axe to get to the stairs, skipping a bunch of rooms in the pinnlace. Use the succubus glitch with the cutall to glitch under the stairs on the way back.

I have done a decent Julius mode run, finishing in :49:05. Doesn't really matter as long as nobody runs the normal/new game mode I guess. Can't record it anyway.

let me clarify that i won't be doing this run for a long time. i don't have the proper equipment either.

Nitrodon -- your route seems pretty solid. I'm just skeptical about all that weapon synthesizing. it seems it would require a lot of luck and frustration. but you may know more about the game than me.

Altair -- Barbariccia isn't going to work. it eats up MP really quickly and won't save too much time in the longrun. for NewGame+, its a definite.

XtraX -- before I read the whole thing, i was ready to say "WHAT THE HELL DID I JUST SAY!!!???" but actually that seems very different than what the Succubus glitch is actually used for. interesting, but that brings us back to luck and frustration. Succubus is a 3 star rarity, isnt it?
I have no solution, but i admire the problem.

Hmm, so this run will never be up at SDA. Don't you know somebody near you who has a good-quality digital camera? or don't you have a DS lite?

Anyways, it would be nice if you would upload this run just for the forum. You could simply record your test runs with a decent camera and when you stop running this you upload the best one. I think every DoS player wants to see a DoS run, even it's not that good.

i do have a DS Lite. just not a digital camera.  i'll see what i can do though.

::edit:: about Julius mode, which bosses must be beaten? is it like Soma's game where Aguni is the required boss? cause i was led to believe that all the bosses have to be beaten in Julius mode.
You need a decent weapon in order to kill Zephyr quickly.  At the level you will naturally reach before the fight, the Cutlass only deals 5 damage to him, which requires you to hit him 247 times.  The Fragarach deals 12-13 damage instead, requiring only 95-103 hits.  I prefer swords (since I haven't mastered the backdash cancel), so I haven't tested other types of weapons.

The run is segmented such that each segment only contains one randomly dropped soul.  This helps to minimize frustration in manipulating souls.  The most frustrating soul for me to manipulate in this run was probably Killer Clown, which is a required soul.

Well, look at the new AoS topic ... they're going to buy Mind ups for Manticore, remember you're also invincible while transformed, and Barbariccia doesn't need as much space as Manticore even though the sprite is bigger. IIrc, there's a Barbariccia near the first Dario fight, and there's a save room as well.
The Backdash Cancel is quite simple. Attack, Backdash, immediatly tap down, repeat. If you hold the direction you are facing between the tapping down and reattacking part you can even move forwards during it.

i tried a playthrough last night. i used the great swords throughout the game. in other words, once i got the Claymore, i equipped it and upgraded it to Falchion with a Warg. then, after Malphas, i got a Great Armor and upgraded to the Great Sword. kept that until Durandal came around.

my time came to 1:18, but that involved less than perfect luck and a detour around the chapel. i accidently sold the Cutall and didn't notice.

concerning New Game +, i believe that right from the Wizardry Lab, you can detour to Subterranean and then to the Ruins. Get Bat Company so that Malphas (and Balore i think) can be skipped. Go to the garden, kill Dario, backstab to Clock Tower, etc. have to check that though

and I'm clueless on Julius mode.

IIrc, there are two ways to Silenced Ruins: The initial entrance requires Zephyr soul, and the backdoor can't be reached w/o double jump, I think.

You can go straight to Dario, defeat him and get Alucard. After that you can pretty much go wherever you want. IIrc, you have to kill every boss except Paranoia to open the entrance to Mine of Judgement. However, you'll probably want to go get Yoko first, she will speed up many boss battles.

You need the proper seal for Bat Company, so that won't be possible in the first place.

I just did a really, really horrible (:D) NG+ test run. The time at the final save before Menace was about 40 minutes, so .... I don't know. 10 minutes of mistakes? Not really. Would it make sense to aim for sub-35?

Oh, btw, Valkyrie is faster than Barbariccia.

EDIT: Yorick is definitely worth getting. Equip it, do a tiny jump, press up + B and slide into the skull. Heavy damage. Useful for Zephyr, who took me like 6 minutes at level 13. >_>

just tried a test for NG+. my final time was about 41 minutes. there were three missed seals though and a couple little mishaps. i think sub40 is what to aim for there.

my initial setup was Erinys, Valkyrie, Lilith, Valmanway, Death's Robe, and Chaos Ring. That stays the same throughout most of the game.

for the bosses:
Flying Armor: couple Erinys blasts
Balore: Abaddon first, Valkyrie FTW
Dmittri: Erinys with Valmanway in between
Malphas: charge into him with Valkyrie, Erinys FTW
Dario: Erinys and Claimh Solias special attack, Frozen Shade would probably be a good idea.
Zephyr: Aguni
Bat Company: Aguni
Little Paranoia: Valkyrie into a wall
Big Paranoia: Erinys, maybe Abaddon
Aguni: Erinys and Medusa Head
Death: Erinys, Medusa Head if 2nd form does the skull thing
Abaddon: Aguni or Erinys
Menace: Erinys and Medusa Head
Stolas is better than Lilith.

actually, i remember something on Game FAQs mentioning infinite Killer Clown with the Werewolf. i think it was Advanced Boss Rush strategy or something like that.

Okay, now I actually tried Yorick. Amazing. Deals about 180 damage to Zephir for 20 mana. I also got Mandragora, mainly for those annoying Mini Devils / Imps / whatever in Clock Tower, but it's also nice if you're surrounded by enemies.
