i have a few questions to ask about this game.

1: how much faster then the american version is the JP? the only thing im certain about is that it only takes 2 ganado kills in the village to make the ''reinforcements'' come in instead of 5.

2: is it possible to get sub-2:00, given the load times?

3: if i were to submit a time+video to this site of a run, would it be legal to cut out the loading screens?

4: what are the best weapons to use on a new game+ speed run? my inventory is:
tommy gun
handcannon
IRL
8 of each kind of grenade
6 g+r herb

I'm pretty sure it's policy to leave the loading times in for any game.  I have no idea about high level RE4, but I hope I was helpful in this at least.
Are you talking about the Japanese PS2 version or the Japanese GC version? 

I haven't seen any videos of the Japanese version PS2 village fight, so I can't confirm that about only having to kill 2 ganados on their ps2 version.  But I know that's the case on the Japanese GC, and PAL normal-difficulty GC versions.

Yeah, you won't be able to cut out any loading screens for the run.  Especially since the game has an in-game timer.

As for weapons, you'll definitely want more flash grenades.  You can use 7 or 8 in 5-2 alone.  If you're using them in the 1st half of the water room, that'll be another 3-4 depending on the ganados.

There have been times submitted to Shin's boards of 1:55:xx on the ps2.

I don't see much use for the handcannon.  The only real use that it has is a slightly faster running time in 2-3, and shooting El Gigante #2's doors down a little faster.  But you have to switch weapons too.  And Pota got his 1:49:25 using the IRL for both doors, all the running, and all the way to the first part of the water room in 3-1, so I don't think the handcannon makes much of a difference.

As for hand grenades, obviously the usual places are the 2 groups of ganados in the village fight, the trap/cabinet at the end of 1-1, and sometimes one for the middle machine gunner on the ground in 5-4.  So you probably don't need 8 unless you have a special strategy for them.

You can kill Mendez faster with Handcannon. But it's not much use anyway. I got 1-2 seconds faster times with the Handcannon Tongue
Yeah, that's true.  I guess my aim with it probably isn't good enough to finish Mendez quicker.

Something that would be interesting to exploit is Ashley's knight costume.  I know you don't have to push any of the dodge prompts in 3-4 with it.  That saves the long cutscene of her falling down and getting up on each try.

Sarou:  I'm curious, when you were recording your run, were the times for each time you did a chapter (when you were practicing it) pretty close together?  Ie. Within a second, or were you getting 3-4 second improvements for a level?

I know I was trying to improve my 1-2 time (not recording or anything--just trying to get a good speed run time to post on Shin's board,) and it took what seemed to be a lot of time just to get a one second improvement.  (my best time for 1-1 is 3:50, and 1-2, 6:39.)

I'm just wondering if I'm missing something really big (ie. if other speed runners are going from 6:42 to 6:39 more quickly than I am.  Ie. in 3 second improvements, versus a one-second improvement over 3 hours of play.)

well it depends on the level. For the simple levels it was 1-2 seconds better or probably nothing. For levels like Del Lago or Cabin Fight, it can be something like 15 seconds. I usually played the level first without recording until it's "perfected" and then tried to beat it while recording, so... But sometimes my friend Randomness didn't want me to do so.
